Ours is the first laboratory to succeed in routinely culturing nonpigmented ciliary epithelium (NPE). The graph shows results from an experiment using a monolayer of cultured porcine NPE. Fluorescein-labelled dextran placed on one side of the monolayer, either on the apical side (pink symbols) or basolateral side (yellow symbols) was unable to diffuse across the cell barrier. Cultured cells never behave in precisely the same way as the native cells from which they were derived. Nevertheless, the cultured NPE cells express many of the same proteins found in the native NPE. We are happy to share the cell culture technique with you if you are interested.
To study the regulation of the ion transport mechanisms that drive aqueous humor secretion we can combine results from:
1. Studies of transport across cultured porcine NPE monolayers
2. Studies of transport across the intact porcine iris-ciliary body
3. Studies of aqueous formation by the intact, perfused, porcine eye
4. Biochemical studies on molecules isolated from fresh porcine ciliary epithelium
